چکیده

The orientation of the biological molecule immobilized on a solid surface has been critical in development of various applications. In this study, orientation of antibody was retained by protecting the antigen-binding site of the antibody prior to immobilization to -functionalized mixed self-assembled monolayer (SAM) of 12-mercaptododecanoic acid and 1-heptanethiol. More importantly, the number of immobilization bonds formed between each antigen-binding site protected antibody molecule and the solid surface was controlled by optimizing the mole fraction of the activated carboxyl group of the linker molecules in the mixed SAM. The amount of antibody used in this study was approximately equivalent to the amount for one monolayer surface coverage. The resulting activity of protected immobilized antibody was about 10 fold higher than that of random immobilized antibody.

Development of Non-Labeled QCM Biosensor for the Detection of β-Galactosidase: A Comparative Study of Gold and Polystyrene Nanoparticles

The performance of gold and polystyrene nanoparticles was investigated using quartz crystal microbalance (QCM) as sensor platform; β-Galactosidase antibody with corresponding antigen was utilized for the immunoreaction. The development of the immunosensor included: 1) formation of self assembled monolayers on quartz crystals; 2a) immobilization of p-aminothiophenol functionalized gold nanoparti...

Selective immobilization of proteins to self-assembled monolayers presenting active site-directed capture ligands.

This paper describes a method for the selective and covalent immobilization of proteins to surfaces with control over the density and orientation of the protein. The strategy is based on binding of the serine esterase cutinase to a self-assembled monolayer presenting a phosphonate ligand and the subsequent displacement reaction that covalently binds the ligand to the enzyme active site. Surface...

Fabrication of antibody arrays using thermally responsive elastin fusion proteins.

A general method has been developed to immobilize antibodies onto an array surface by employing fusion proteins consisting of an elastin domain with tunable hydrophobic properties and an antibody-binding domain with high binding affinity and specificity for antibodies. Antibodies conjugated with the elastin fusion proteins can be directly printed on a self-assembled monolayer-modified glass sli...

Label Free QCM Immunobiosensor for AFB1 Detection Using Monoclonal IgA Antibody as Recognition Element

This study introduces the use of an IgA isotype aflatoxin (AF) specific monoclonal antibody for the development of a highly sensitive Quartz Crystal Microbalance (QCM) immunobiosensor for the detection of AF in inhibitory immunoassay format. The higher molecular weight of IgA antibodies proved an advantage over commonly used IgG antibodies in label free immunobiosensor measurements. IgA and IgG...

Design and characterization of a metal ion-imidazole self-assembled monolayer for reversible immobilization of histidine-tagged peptides.

We report the design and characterization of a metal ion-imidazole self-assembled monolayer on a gold electrode. The resultant monolayer is well-suited for direct immobilization of histidine and methylene blue-modified peptides.
